A claim contesting her virginity means that he married her on the assumption that she was a virgin but the wedding night experience did not support that claim, i.e., there was neither bleeding nor tightness.
Hilchos Ishus 11:10
If a man marries a virgin with a kesubah of 200 and claims not to find the signs of virginity, the woman is questioned. If she says that her hymen was damaged by a fall onto a piece of wood or the ground, she is believed and her kesubah is adjusted to 100 zuz. Even if the husband suspects that she engaged in relations and feels that he should not be responsible for her, his claim is not accepted because it is mere speculation on his part.
